S-898270 is a selective phosphodiesterase 4D (PDE4D) inhibitor being developed by Shionogi & Co., Ltd. for the treatment of cognitive impairment and the enhancement of memory formation. By inhibiting the PDE4D enzyme, the drug aims to increase intracellular levels of cyclic adenosine monophosphate (cAMP) in the brain, which is a critical secondary messenger for synaptic plasticity and long-term potentiation. This mechanism is intended to improve cognitive function in patients with neurodegenerative conditions such as Alzheimer's disease. In late 2024, Shionogi initiated a Phase 1 clinical trial in Japan, partnering with Cumulus Neuroscience to utilize the NeuLogiq® digital health platform for the collection of objective neurophysiological data, including EEG and cognitive biomarkers, in a home-based setting.
