S-adenosyl-L-methionine + betaine is a fixed-dose combination of two methyl donors, S-adenosyl-L-methionine (SAMe) and betaine. SAMe is a naturally occurring compound in the body involved in methylation reactions, neurotransmitter synthesis, and maintenance of cell membranes. Betaine acts as a methyl group donor and helps regulate homocysteine metabolism. The combination has been studied primarily for its potential to enhance antidepressant effects in patients with mild-to-moderate depression who are low responders to conventional antidepressants. Betaine may also counteract the increase in homocysteine sometimes seen with SAMe therapy and can raise plasma levels of SAMe, potentially potentiating its effect[5]. Preclinical studies suggest that this combination may have hepatoprotective effects by maintaining glutathione homeostasis under oxidative stress or ethanol-induced liver injury[4]. The product DDM Metile is an example of this fixed-dose combination used clinically for depressive symptoms[5].
