**s-amlodipine + olmesartan** is a fixed-dose combination drug containing s-amlodipine, a dihydropyridine calcium channel blocker, and olmesartan medoxomil, an angiotensin II receptor blocker[2][3][1]. The combination is indicated for the treatment of hypertension, particularly in cases where monotherapy is ineffective. Mechanism of action: s-amlodipine inhibits L-type calcium channels in vascular smooth muscle, leading to vasodilation and reduction of blood pressure; olmesartan selectively blocks the angiotensin II type 1 receptor, inhibiting the effects of angiotensin II, subsequently reducing vasoconstriction and aldosterone-mediated volume expansion[2][3][1]. Clinical studies have shown that the combination is more effective at lowering blood pressure than either drug alone, with improved rates of achieving guideline blood pressure goals and a side-effect profile that is generally favorable and similar to the individual constituents[1][3]. Primary indication is the management of hypertension as monotherapy or in combination with other antihypertensive agents. The combination is available as a generic and as branded formulations, e.g., Azor (USA)[2][3]. The product is developed and marketed by Daiichi Sankyo[3] and was approved for use in the US in September 2007[2].
