S-amlodipine gentisate is a crystalline acid addition salt formed by reacting the active (S)-enantiomer of amlodipine with gentisic acid. It is developed to provide improved potency and safety compared to racemic amlodipine formulations. S-amlodipine acts as a calcium channel blocker of the dihydropyridine class, selectively inhibiting L-type calcium channels in vascular smooth muscle cells more than in cardiac muscle cells. This results in vasodilation, reduced peripheral resistance, and lower blood pressure. The addition of the gentisic acid moiety aims to enhance stability and maintain effective blood concentrations for longer durations after administration. S-amlodipine (levamlodipine) is indicated for hypertension and cardiovascular diseases such as angina[3][5][6][7].
