S-FLU is an investigational **single-cycle influenza vaccine** designed as a replication-deficient, pseudotyped influenza A virus. In the original Oxford-described construct, suppression or replacement of the influenza hemagglutinin coding function prevents productive replication, while the virion is coated with functional hemagglutinin supplied in producer cells, allowing only a single round of infection. This enables delivery and expression of conserved internal influenza antigens such as nucleoprotein and matrix proteins, with the aim of inducing broad heterosubtypic cellular immunity rather than primarily strain-specific neutralizing antibody responses. The platform was developed by researchers at the University of Oxford and has been studied preclinically, including inhaled or mucosal administration in animal models, as a potential broadly protective or universal influenza A vaccine candidate.
