S-Gboxin is a small molecule and a functional analogue of Gboxin, designed as an inhibitor of oxidative phosphorylation (OXPHOS). It specifically inhibits the growth of mouse and human glioblastoma (GBM) cells by targeting mitochondrial energy metabolism. The primary mechanism involves inhibition of the F1F0-ATPase (also known as ATP synthase), which is essential for ATP production in mitochondria. S-Gboxin demonstrates potent antitumor activity in preclinical models, with an IC50 against GBM cells around 470 nM. It is being investigated primarily for its potential to enhance standard therapies for glioblastoma and diffuse midline glioma[1][2][4][7].
