s-KL (secreted Klotho) is a secreted isoform of the alphaKlotho protein, an aging suppressor. It is a 70 kDa protein that exerts pleiotropic beneficial properties by modulating insulin, insulin-like growth factor-1 (IGF-1), and Wnt signaling pathways, while also reducing inflammatory and oxidative stress. It has demonstrated therapeutic potential in preclinical models for enhancing remyelination by oligodendrocytes in multiple sclerosis, improving physical fitness, bone microstructure, and cognitive functions in aging, and inhibiting osteogenic differentiation in renal interstitial biomineralization. Delivery methods explored include adeno-associated virus (AAV) vectors and mesenchymal stem cells (MSCs) transduced with s-KL.
