Drug intelligence / Profile preview
S-p-bromobenzylglutathione cyclopentyl diester (BBGC, also known as BrBzGCp2) is a cell-permeable, selective small molecule inhibitor of glyoxalase 1 (GLO1). GLO1 is a rate-limiting enzyme in the glyoxalase system responsible for detoxifying methylglyoxal (MG), a reactive dicarbonyl byproduct of glycolysis. By inhibiting GLO1, BBGC induces the accumulation of cytotoxic methylglyoxal, leading to dicarbonyl stress, activation of stress-activated protein kinases (such as JNK1 and p38 MAPK), and subsequent apoptosis. BBGC has been widely utilized as a preclinical research tool to investigate the role of GLO1 and methylglyoxal in cancer biology, neurobiology (including anxiety and epilepsy), and metabolic diseases such as diabetes.
