Overview

S-primaquine is the (+)-enantiomer of the 8-aminoquinoline antimalarial drug primaquine. While primaquine is clinically administered as a racemic mixture, the S-enantiomer exhibits distinct pharmacokinetic and toxicological properties. Research indicates that S-primaquine is typically cleared more rapidly than the R-enantiomer and may be more closely associated with the drug's characteristic hemolytic toxicity in individuals with glucose-6-phosphate dehydrogenase (G6PD) deficiency. Like the racemate, it is studied for its ability to eliminate hepatic hypnozoites of *Plasmodium vivax* and *Plasmodium ovale* (radical cure) and its gametocytocidal activity against *Plasmodium falciparum*.
