Drug intelligence / Profile preview
S-SK609 is a G-protein biased signaling agonist of the dopamine D3 receptor (D3R) and an inhibitor of the norepinephrine transporter (NET). It is the active S-enantiomer of the racemic compound SK609. Developed by Drexel University and licensed to PolyCore Therapeutics, S-SK609 is designed to treat motor symptoms and levodopa-induced dyskinesia (LID) in Parkinson's disease, as well as cognitive impairment. Unlike traditional dopamine agonists, S-SK609 selectively activates D3R without recruiting beta-arrestin-2, thereby avoiding receptor desensitization, tolerance, and dyskinesia. Additionally, its selective inhibition of NET increases dopamine and norepinephrine levels in the prefrontal cortex, promoting cognitive enhancement without the psychostimulant-like side effects associated with dopamine transporter (DAT) inhibition.
