**S.Br@DBCO** is an experimental orally administered live-biotherapeutic construct consisting of the probiotic yeast *Saccharomyces boulardii* surface-modified with dibenzocyclooctyne. It was developed as part of a bioorthogonal, inflammation-targeted strategy for inflammatory bowel disease: S.Br@DBCO is intended to bind azide-modified Prussian blue nanozyme at inflamed intestinal sites through copper-free azide-DBCO click chemistry. In a dextran sulfate sodium mouse colitis model, the paired PB@N3 plus S.Br@DBCO system increased intestinal probiotic retention, restored intestinal-barrier function, reduced intestinal inflammation, and improved colitis-associated outcomes.
