S066913 (also known as XEN-D0103) is a highly selective small molecule inhibitor of the atrial-specific potassium current IKur, which is mediated by the Kv1.5 channel. Developed by Servier in collaboration with Xention, it was investigated as a potentially safer antiarrhythmic treatment for paroxysmal atrial fibrillation (PAF) due to the absence of Kv1.5 channels in the ventricles, thereby minimizing the risk of ventricular pro-arrhythmia. The drug was evaluated in the Phase II DIAGRAF-IKUR trial, which utilized insertable cardiac monitors to assess AF burden. However, the trial was terminated prematurely due to non-study-related preclinical safety concerns, and the drug failed to show clinically meaningful efficacy at the tested doses (5 mg to 100 mg once daily).
