Overview

S095029 is a clinical-stage monoclonal antibody that targets NKG2A (killer cell lectin-like receptor C1), an inhibitory receptor expressed on natural killer (NK) cells and some T cells. By blocking NKG2A, S095029 is designed to enhance anti-tumor immune responses by preventing the interaction between NKG2A and its ligand HLA-E. This blockade releases the inhibition of NK and CD8+ T cells, potentially increasing their cytotoxic activity against tumor cells. The antibody has been engineered with an Fc-silenced backbone to minimize unwanted immune activation through Fc-mediated effector functions[9][5]. It is being developed primarily for use in combination with other immunotherapies such as pembrolizumab (anti-PD-1) or cemiplimab (anti-PD-1), particularly in advanced solid tumors including metastatic non-small cell lung cancer and gastroesophageal junction/gastric adenocarcinoma[4][6][8]. The drug was originally developed by Adir SRL and Symphogen (a subsidiary of Servier), with current development led by Servier Bio-Innovation LLC[5].
