S095033 (also known as AG-270) is an oral small molecule inhibitor of methionine adenosyltransferase 2A (MAT2A), the enzyme primarily responsible for the synthesis of S-adenosylmethionine (SAM). MAT2A inhibition targets cancers with homozygous deletion of MTAP or loss of MTAP protein, which occurs in approximately 15% of cancers. By inhibiting MAT2A, S095033 reduces SAM levels and disrupts methylation-dependent processes critical for tumor cell survival. The drug has shown preliminary clinical activity in phase 1 trials as monotherapy in patients with advanced malignancies harboring MTAP deletions or loss[1][3][4]. It is being developed by Agios Pharmaceuticals and Servier for various solid tumors including adenocarcinoma and squamous cell cancer[2]. Common treatment-related toxicities include reversible increases in liver function tests, thrombocytopenia, anemia, and fatigue. Partial responses and stable disease have been observed in early clinical studies[1][3].
