S100A4-11 (also referred to as S1004-11) is a humanized monoclonal antibody designed to target and neutralize extracellular S100 calcium-binding protein A4 (S100A4). Developed by researchers at the Houston Methodist Research Institute, the Texas Therapeutics Institute, and Baylor College of Medicine, S100A4-11 acts as a functional blocking antibody to inhibit the metastasis-promoting and immunosuppressive activities of soluble S100A4. By neutralizing extracellular S100A4, the antibody blocks the formation of the premetastatic niche (PMN), suppresses neutrophil infiltration, and reverses the immunosuppressive tumor microenvironment, thereby activating natural killer (NK) and T cells. S100A4-11 is being investigated in preclinical models for the prevention and treatment of cancer metastasis, particularly in breast cancer and glioma.
