The S100A4-TfR bispecific antibody (BsA) is a preclinical-stage biologic engineered to treat glioblastoma (GBM) by overcoming the blood-brain barrier (BBB). It features a dual-targeting mechanism: one arm binds to S100A4 (Protein S100-A4), a protein that regulates stemness, epithelial-mesenchymal transition (EMT), and immunosuppressive phenotypes in the tumor microenvironment; the second arm targets the Transferrin Receptor (TfR1) to facilitate robust BBB penetration through receptor-mediated transcytosis. By blocking S100A4, the antibody aims to reprogram myeloid and T cells toward a pro-inflammatory state, increase CD4+ T cell infiltration, and reduce the self-renewal capacity of glioma stem cells. It is currently being developed by the University of Texas Health Science Center at Houston and the Houston Methodist Research Institute.
