Overview

S107 is a small molecule benzothiazepine derivative that acts as a stabilizer of the ryanodine receptor 2 (RyR2) and FK506-binding protein 12.6 (FKBP12.6/calstabin2) complex. It belongs to a class of compounds known as "Rycals." By stabilizing this interaction, S107 prevents the pathological leak of calcium from the sarcoplasmic reticulum into the cytosol, a process implicated in various conditions including heart failure, catecholaminergic polymorphic ventricular tachycardia (CPVT), and Duchenne muscular dystrophy. Research also indicates its potential in treating pulmonary hypertension by inhibiting hypoxic pulmonary artery remodeling and vasoconstriction. S107 was developed through research at Columbia University and ARMGO Pharma as a prototype therapeutic candidate.
