Overview

S14161 is a small molecule inhibitor of the phosphoinositide 3-kinase (PI3K) signaling pathway, originally identified through a chemical library screen for compounds that inhibit D-cyclin expression. By targeting PI3K, S14161 effectively blocks the downstream activation of the AKT/mTOR cascade, which is a critical driver of cell survival, proliferation, and metabolism in many cancers. Preclinical studies have demonstrated that S14161 induces apoptosis and inhibits growth in multiple myeloma, leukemia, and prostate cancer cells. It also appears to modulate the epithelial-mesenchymal transition (EMT) and can sensitize cancer cells to other chemotherapeutic agents like cisplatin. S14161 is primarily utilized as a research tool in preclinical oncology studies to investigate the role of the PI3K/AKT/mTOR pathway in tumor progression and resistance.
