Overview

S2383 is a potent and selective small molecule inhibitor of 11-beta-hydroxysteroid dehydrogenase type 1 (11β-HSD1), an enzyme that catalyzes the conversion of inactive cortisone into active cortisol within specific tissues such as the liver, adipose tissue, and brain. Developed by Servier, the compound was primarily investigated for its potential to treat metabolic disorders, including type 2 diabetes and obesity, by reducing local glucocorticoid action and thereby improving insulin sensitivity and glucose metabolism. Furthermore, S2383 was evaluated for its neuroprotective properties, aiming to mitigate cognitive decline associated with Alzheimer's disease and aging by lowering brain cortisol levels, which are known to contribute to hippocampal dysfunction and memory impairment. Clinical development reached Phase 1 trials for both metabolic and cognitive indications before the program was discontinued.
