Drug intelligence / Profile preview
S293 is a peptide mimetic of the C-terminal regulatory site of connexin-36 (Cx36) designed to competitively inhibit cytokine-mediated Cx36 phosphorylation. In diabetes, pro-inflammatory cytokines and free fatty acids disrupt Cx36 gap junction coupling in pancreatic beta-cells, leading to a loss of synchronized calcium dynamics and impaired insulin secretion. By preventing Cx36 phosphorylation, S293 rescues gap junction permeability, restores coordinated calcium oscillations, and protects beta-cells from cytokine-induced apoptosis.
