Overview

S32504 is a novel naphtoxazine small molecule compound acting as a high-efficacy agonist at dopamine D2 and D3 receptors, with preferential activity at D3 over D2. It was developed as an antiparkinsonian and antidepressant agent. Preclinical studies demonstrated pronounced antiparkinsonian effects in rodent and primate models, with rapid onset and durable activity, and without significant dyskinesia. S32504 also exhibited robust neuroprotective effects against dopaminergic neurotoxins in vitro and in vivo, and displays antidepressant-like activity in animal models. Its antiparkinsonian effect is primarily mediated by activation of dopamine D2 receptors, while engagement of D3 receptors contributes to neuroprotection. S32504 was more potent than other preferential D3 agonists such as pramipexole and ropinirole in behavioral and neuroprotective models. Development for major depressive disorder was terminated before approval.
