Overview

S33084 is a **novel, potent, and highly selective antagonist of the dopamine D3 receptor**. It is a small molecule of the benzopyranopyrrole class and exhibits over 100-fold selectivity for dopamine D3 receptors compared to D2 and other receptors[2][5][9]. In various preclinical studies, S33084 has been shown to antagonize dopamine-induced signaling at D3 receptors, block D3 agonist-induced effects such as hypothermia and penile erections, and enhance certain cognitive behaviors in animal models[1][2][3][5]. It does not display antipsychotic or extrapyramidal activity in standard animal models, indicating a distinct behavioral pharmacology compared to typical D2 antagonists[1][2]. S33084 has shown some potential antiparkinsonian effects in rat models, particularly by attenuating bradykinesia, though it does not ameliorate L-DOPA-induced dyskinesia[3]. Developed by Servier, its highest known development phase is discontinued before entering clinical use[8].
