Overview

S47445 (also known as CX1632) is a small molecule positive allosteric modulator (PAM) of the alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) receptor. It was developed by Institut de Recherches Internationales Servier in collaboration with Cortex Pharmaceuticals (now RespireRx Pharmaceuticals) for the treatment of neurodegenerative and psychiatric disorders. The drug's primary mechanism involves enhancing glutamatergic neurotransmission by modulating AMPA receptors, which are critical for synaptic plasticity and cognitive function. S47445 was specifically investigated in Phase 2 clinical trials for patients with mild to moderate Alzheimer's disease who also exhibited depressive symptoms. However, development for Alzheimer's disease was discontinued in 2017 after the drug failed to demonstrate significant efficacy over placebo in a large Phase 2 study.
