Drug intelligence / Profile preview
S55 is a novel small molecule transient receptor potential (TRP) channel activator developed to treat neglected tropical diseases caused by parasitic flatworms. It acts as a dual agonist of the transient receptor potential melastatin praziquantel channel (TRPMPZQ) orthologs in both *Schistosoma mansoni* (Sm.TRPMPZQ) and *Fasciola hepatica* (Fh.TRPMPZQ) with submicromolar potency. Unlike praziquantel, which is ineffective against *Fasciola* species due to structural variations in their TRPMPZQ binding pocket, S55 is designed to tolerate these variations. Ex vivo, S55 induces rapid contraction and tegumental damage in both *S. mansoni* and triclabendazole-sensitive or triclabendazole-resistant *F. hepatica*. S55 has demonstrated a favorable safety profile in HepG2 assays and is active and well-tolerated in vivo.
