S868 is a small molecule agonist of the G protein-coupled receptor 119 (GPR119), developed by Servier for the treatment of type 2 diabetes mellitus. GPR119 is a G protein-coupled receptor primarily expressed in pancreatic beta cells and intestinal enteroendocrine cells (L-cells). Activation of GPR119 by S868 stimulates the glucose-dependent secretion of insulin and the release of incretin hormones, such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). This dual mechanism of action is designed to improve glycemic control and potentially preserve beta-cell function. S868 reached Phase 1 clinical trials, where its safety, tolerability, and pharmacodynamics were evaluated in healthy volunteers and patients with type 2 diabetes. However, like many other GPR119 agonists, its clinical development appears to have been discontinued, likely due to insufficient efficacy in human subjects compared to preclinical models.
