S969 is an oral, mutation-agnostic small molecule multi-kinase inhibitor being developed by Sarcogent Therapeutics for the treatment of Duchenne muscular dystrophy (DMD). It functions by transcriptionally upregulating α7β1 integrin, a structural protein in skeletal and cardiac muscle that serves as a functional surrogate for missing dystrophin. By enhancing α7β1 integrin levels, S969 stabilizes the sarcolemma and protects muscle fibers from damage. Preclinical data in mdx mice and Golden Retriever Muscular Dystrophy (GRMD) canine models have demonstrated that S969 treatment leads to rapid and significant reductions in serum biomarkers of muscle damage, such as creatine kinase (CK) and transaminases (AST/ALT), while improving muscle force, physical activity, and preserving cardiac ejection fraction. As of 2026, the compound is in IND-enabling studies.
