SA-4-1BBL is a novel recombinant fusion protein and CD137 (4-1BB) agonist designed for cancer immunotherapy and immunoprevention. The molecule consists of the extracellular domain of the 4-1BB ligand (4-1BBL) fused to a modified core of streptavidin (SA). This unique streptavidin-based platform allows the protein to form stable tetramers and higher-order oligomers, which are essential for effectively cross-linking the CD137 receptor on T cells and natural killer (NK) cells. Unlike many agonistic monoclonal antibodies targeting CD137, SA-4-1BBL has demonstrated a superior safety profile in preclinical models, avoiding the systemic inflammatory toxicities and hepatotoxicity often associated with this target. It is being investigated for its ability to enhance antitumor immunity and serve as an immunopreventive agent in high-risk populations for cancers such as breast and lung cancer.
