SA-5

SA-5 is a potentially novel, liver-targeted, orally available Toll-like receptor 7 (TLR7) agonist. It is being investigated as an immunostimulatory agent for the treatment of chronic infections and cancer, with the goal of overcoming systemic toxicity challenges often associated with other TLR7 agonists. Preclinical studies in nonhuman primates demonstrated that SA-5 induces dose-dependent type I interferon production with limited systemic inflammation. It exhibited immunostimulatory activity comparable to GS-9620 but with reduced adverse biomarker shifts. Furthermore, in an immunocompetent mouse model of chronic hepatitis B, SA-5 showed the capacity to revitalize impaired exhausted T cells and liver-recruited macrophages, suggesting its potential in reinvigorating antiviral immunity.
