Overview

SA012 is an N-acetylgalactosamine (GalNAc)-conjugated small interfering RNA (siRNA) therapeutic developed by Siran Biotechnology for the treatment of chronic hepatitis B (CHB). It utilizes the company's proprietary eSAFE chemical modification technology and STORK delivery platform to target the mRNA of the programmed death-ligand 1 (PD-L1) gene specifically in liver tissue. By silencing PD-L1 expression, SA012 aims to reverse T-cell exhaustion and enhance the host's antiviral immune response against the hepatitis B virus (HBV), representing a novel immunomodulatory approach to achieving a functional cure. Core preclinical data for SA012 was presented at the EASL 2024 conference.
