Overview

SA4Ag (PF-06290510) is an investigational 4-antigen *Staphylococcus aureus* vaccine developed by Pfizer for the prevention of invasive staphylococcal infections, particularly in high-risk surgical patients. The vaccine is a biologic composed of four key antigens: capsular polysaccharide (CP) serotypes 5 and 8, each conjugated to the CRM197 carrier protein, a recombinant mutant form of clumping factor A (rmClfA or Y338A-ClfA), and a recombinant manganese transporter protein C (rMntC). This multi-antigen approach is designed to induce a broad immune response targeting both the protective capsule and surface proteins essential for bacterial adherence and nutrient acquisition. SA4Ag was granted Fast Track designation by the FDA in 2014 and was evaluated in the Phase 2b/3 STRIVE efficacy study in patients undergoing elective spinal fusion surgery.
