Overview

Sabatolimab is a humanized IgG4 monoclonal antibody that targets T cell immunoglobulin and mucin domain-containing protein 3 (TIM‑3/HAVCR2). It was developed as an investigational first-in-class immuno-myeloid therapy for hematologic malignancies and some solid tumors. By binding to TIM‑3 on immune cells and leukemic stem cells, sabatolimab blocks the immune-suppressive functions of TIM‑3 and enhances Type 1 T helper cell responses including interferon-gamma production. The drug was primarily investigated in myelodysplastic syndromes (MDS), acute myeloid leukemia (AML), chronic myelomonocytic leukemia (CMML), myelofibrosis, glioblastoma multiforme, gliosarcoma and other solid tumors. Despite initial promise—including orphan drug designation for MDS—development was discontinued after pivotal trials failed to meet primary endpoints[1][5][6][8].
