Sabatolimab + spartalizumab is a combination of two investigational monoclonal antibodies developed for the treatment of advanced solid tumors, particularly melanoma and non-small cell lung cancer (NSCLC). Sabatolimab (MBG453) targets T-cell immunoglobulin and mucin-domain containing-3 (TIM-3), an immune checkpoint receptor involved in T-cell exhaustion, while spartalizumab (PDR001) targets programmed death-1 (PD-1), another key immune checkpoint. By blocking TIM-3 and PD-1 pathways, this combination aims to enhance anti-tumor immune responses in patients who have progressed following prior anti-PD-1/PD-L1 therapy. Clinical studies have shown that the regimen is generally well tolerated with limited antitumor activity observed; recommended phase 2 dosing is sabatolimab 800 mg every 4 weeks plus spartalizumab 400 mg every 4 weeks[1][2][3][6].
