Overview

Sabin OPV2 is a live-attenuated oral poliovirus vaccine of type 2, derived from the Sabin strain. It was historically used for preventive immunization against poliomyelitis type 2 and later limited to emergency outbreak response following the global cessation of routine type 2 OPV use in 2016. While effective in inducing immunity, a significant limitation of Sabin OPV2 is its potential to revert to neurovirulence and cause circulating vaccine-derived poliovirus type 2 (cVDPV2) outbreaks, particularly in under-vaccinated populations. This genetic instability led to the development of more genetically stable novel OPV2 (nOPV2) vaccines, which are now replacing Sabin OPV2 for outbreak responses.
