**Sabutoclax** is a pan-active small molecule antagonist of the BCL-2 family of anti-apoptotic proteins, acting as a BH3 mimetic. It binds to and inhibits several key anti-apoptotic BCL-2 family proteins including BCL-2, MCL-1, BCL-xL, and BFL-1. By antagonizing these proteins, sabutoclax induces apoptosis in tumor cells, enhances chemotherapy sensitivity, eliminates cancer stem cells, and reduces drug resistance in diverse cancer types such as breast, prostate, leukemia, colorectal, and pancreatic cancer. Its activity includes down-regulating the IL-6/STAT3 pathway, which is associated with cancer stem cell maintenance and resistance. Developed from apogossypol derivatives for improved efficacy and selectivity, sabutoclax remains in preclinical development and has shown synergistic effects when combined with chemotherapeutic agents in preclinical models[1][2][3][4][5][7].
