Overview

SAC-253 is a first-in-class antibody-drug conjugate (ADC) developed by Saccharo Inc. that targets de-N-acetyl-polysialic acid (dPSA). dPSA is a unique, highly conserved polysaccharide specifically expressed on the surface of various cancer cells, including pancreatic, lung, breast, and prostate cancers, but is absent in normal tissues. It is typically found linked to the nuclear protein nucleolin on the cell surface and is implicated in tumor immune evasion. SAC-253 is designed to selectively bind to dPSA-expressing tumor cells and deliver a cytotoxic payload, thereby inducing targeted cell death while minimizing off-target effects. The drug is currently in preclinical development.
