SacI-81 is an antibody-drug conjugate (ADC) developed by Corellia AI, utilizing its proprietary Coromip technology to deliver a potent small molecule payload to TROP2-expressing cancer cells. The ADC consists of a monoclonal antibody targeting Trophoblast cell-surface antigen 2 (TROP2) conjugated to omipalisib (compound '81'), a dual inhibitor of Phosphatidylinositol 3-kinase (PI3K) and Mammalian target of rapamycin (mTOR). By specifically binding to TROP2, which is overexpressed in various solid tumors, SacI-81 facilitates the intracellular delivery of omipalisib, thereby inhibiting the PI3K/AKT/mTOR signaling pathway and inducing tumor cell death. SacI-81 is currently in preclinical development for multiple indications, including ER+/HER2- breast cancer, triple-negative breast cancer (TNBC), ovarian cancer, endometrial cancer, and EGFR- or KRAS-mutant non-small cell lung cancer (NSCLC).
