sacituzumab govitecan (Immunomedics)

Sacituzumab govitecan is a Trop-2-directed antibody-drug conjugate developed initially by Immunomedics and subsequently by Gilead. It consists of a humanized anti-Trop-2 antibody linked through a hydrolyzable CL2A linker to SN-38, the active topoisomerase I-inhibitor metabolite of irinotecan. Trop-2-mediated binding and internalization concentrate the conjugate in tumor tissue, where payload release causes topoisomerase I inhibition and DNA damage. It is marketed as Trodelvy for unresectable locally advanced or metastatic triple-negative breast cancer and previously treated hormone receptor-positive HER2-negative metastatic breast cancer.
