Sacituzumab govitecan (IMMU-132) is an antibody-drug conjugate composed of a humanized anti-Trop-2 monoclonal antibody linked to SN-38, the active metabolite of irinotecan and a topoisomerase I inhibitor. The drug targets Tumor-associated calcium signal transducer 2 (Trop-2), a cell surface glycoprotein overexpressed in many epithelial cancers. Upon binding to Trop-2 on tumor cells, the ADC is internalized and releases SN‑38 intracellularly via a moderately stable linker that cleaves at low pH, leading to DNA damage and apoptosis in cancer cells. Sacituzumab govitecan has demonstrated efficacy in multiple solid tumors, particularly metastatic triple-negative breast cancer (mTNBC), and is approved under the brand name Trodelvy for this indication[1][3][6]. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis; it is widely used as part of combination regimens for various solid tumors[4]. The combination of sacituzumab govitecan with carboplatin represents an investigational regimen being explored for synergistic effects in resistant or relapsed cancers.
