Drug intelligence / Profile preview
This is a combination of three anti-cancer agents: sacituzumab govitecan, enfortumab vedotin, and zimberelimab. - **Sacituzumab govitecan** is an antibody-drug conjugate (ADC) targeting Trop-2, delivering the topoisomerase I inhibitor SN-38. It is FDA-approved for metastatic triple-negative breast cancer and under investigation for advanced urothelial carcinoma and other solid tumors[1][5][7]. - **Enfortumab vedotin** is an ADC targeting Nectin-4, with a monomethyl auristatin E (MMAE) payload, approved for metastatic urothelial carcinoma[2][5]. - **Zimberelimab** is a monoclonal antibody targeting PD-1, classified as an immune checkpoint inhibitor; it is investigational in several malignancies. The combination leverages complementary mechanisms: direct cytotoxic targeting of different tumor antigens (Trop-2 and Nectin-4) and immune reactivation via blockade of PD-1. The combination is being studied in advanced/metastatic urothelial carcinoma and other advanced solid tumors.
