Overview

Sacituzumab govitecan + talazoparib is a combination therapy being investigated for metastatic triple-negative breast cancer (mTNBC). This combination leverages the synergistic effects of two distinct mechanisms: sacituzumab govitecan (SG), an antibody-drug conjugate that delivers the topoisomerase 1 inhibitor SN-38 to cancer cells expressing TROP-2, and talazoparib, a poly(ADP-ribose) polymerase (PARP) inhibitor. While concurrent administration of these agents caused intolerable myelosuppression, an innovative sequential dosing schedule has shown improved tolerability and preliminary efficacy. The sequential approach involves administering SG on days 1 and 8, followed by talazoparib on days 15-21 of a 21-day cycle, with growth factor prophylaxis required.
