Sacituzumab govitecan + toripalimab is a combination therapy under investigation for advanced cancers, particularly as a first-line treatment for advanced or metastatic triple-negative breast cancer (TNBC). Sacituzumab govitecan is an antibody-drug conjugate (ADC) that targets TROP-2 on tumor cells; it delivers the topoisomerase I inhibitor SN-38 via a humanized anti-TROP-2 antibody, causing DNA damage and apoptosis in target cells. Toripalimab is a humanized IgG4 monoclonal antibody that acts as an immune checkpoint inhibitor by blocking the programmed cell death protein 1 (PD-1) receptor, reactivating T cells to enhance anti-tumor immune responses. Together, this combination unites targeted cytotoxic activity with immune checkpoint blockade to improve anti-cancer efficacy, with ongoing studies in the first-line setting for TNBC and other malignancies[1][2][4][5][7].
