**Sacituzumab tirumotecan + carboplatin** is an investigational combination therapy composed of sacituzumab tirumotecan (MK-2870, sac-TMT), a novel antibody-drug conjugate (ADC) targeting trophoblast cell surface antigen 2 (TROP2), and carboplatin, a standard platinum-based chemotherapy. Sacituzumab tirumotecan is designed to selectively deliver a topoisomerase I inhibitor payload to TROP2-expressing cancer cells, inducing cytotoxicity by causing DNA damage. Carboplatin forms DNA crosslinks that prevent cell division, enhancing chemotherapy efficacy. This combination is being studied primarily for non-small cell lung cancer (NSCLC) and other solid tumors, typically following progression on prior therapies. The combination's clinical evaluation is ongoing, with phase 1 and 3 studies in various advanced/metastatic solid tumors and NSCLC[1][3][7].
