Overview

**Sacituzumab tirumotecan + enfortumab vedotin** is a combination of two antibody-drug conjugates being investigated for the treatment of advanced or metastatic urothelial carcinoma. Both are designed to selectively deliver cytotoxic agents to cancer cells expressing their respective targets. Sacituzumab tirumotecan (MK-2870) targets the TROP2 antigen and delivers a camptothecin-derived payload (inhibiting topoisomerase I), while enfortumab vedotin targets Nectin-4 and delivers the microtubule-disrupting agent monomethyl auristatin E (MMAE). The combination is currently being studied in early-phase clinical trials for safety and efficacy in bladder and urothelial cancer patients[1][3][5][9][10].
