Sacituzumab tirumotecan (also known as SKB264 or MK-2870) is a TROP2-directed antibody-drug conjugate (ADC) that utilizes a pH-sensitive carbonate linker to attach a potent topoisomerase I inhibitor payload (a belotecan derivative) to a humanized monoclonal antibody. Pucotenlimab (also known as putolizumab or HX008) is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor to restore T-cell mediated anti-tumor activity. The combination aims to leverage the direct cytotoxic effects of the ADC and the immune-potentiating effects of the PD-1 inhibitor to improve outcomes in patients with biliary tract cancers, particularly in studies led by Sir Run Run Shaw Hospital.
