Overview

Sacituzumab tirumotecan + sintilimab is an investigational combination therapy comprising a TROP2-directed antibody-drug conjugate (ADC) and a PD-1 immune checkpoint inhibitor. Sacituzumab tirumotecan (also known as SKB264 or MK-2870) consists of a humanized monoclonal antibody targeting Trophoblast cell-surface antigen 2 (TROP2), which is frequently overexpressed in epithelial tumors, conjugated to a novel topoisomerase I inhibitor payload (tirumotecan) via a stable linker. Sintilimab is a fully human IgG4 monoclonal antibody that binds to the Programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated antitumor immunity. This combination is designed to leverage the cytotoxic effects of the ADC, which can induce immunogenic cell death and increase tumor antigen presentation, to potentially enhance the efficacy of the PD-1 inhibitor. It is primarily being evaluated for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC) and other solid tumors.
