sacituzumab tirumotecan + toripalimab is a combination therapy comprising a trophoblast cell surface antigen 2 (TROP2)-directed antibody-drug conjugate (ADC) and a programmed cell death protein 1 (PD-1) checkpoint inhibitor. Sacituzumab tirumotecan (also known as SKB264 or MK-2870) consists of a humanized anti-TROP2 monoclonal antibody conjugated via a novel linker to a cytotoxic belotecan-derived topoisomerase I inhibitor payload (KL610023), with a drug-to-antibody ratio (DAR) of 7.4. Toripalimab is a humanized IgG4 monoclonal antibody that blocks the interaction between PD-1 and its ligands, PD-L1 and PD-L2, thereby restoring T-cell mediated anti-tumor immunity. This combination is being investigated as a first-line treatment for patients with PD-L1 positive unresectable locally advanced or metastatic triple-negative breast cancer (TNBC), leveraging the potential synergy between ADC-induced immunogenic cell death and immune checkpoint blockade.
