Overview

Saclorin is an immunosuppressant medication containing the active ingredient cyclosporin, manufactured by Pioneer Pharmaceuticals. It is a cyclic nonribosomal peptide that acts as a potent calcineurin inhibitor. The drug's mechanism involves binding to the cytosolic protein cyclophilin A (PPIA) to form a cyclosporin-cyclophilin complex. This complex then binds to and inhibits calcineurin, a calcium-dependent serine/threonine phosphatase. Inhibition of calcineurin prevents the dephosphorylation and subsequent nuclear translocation of the Nuclear Factor of Activated T-cells (NFAT), which effectively blocks the transcription of interleukin-2 (IL-2) and other cytokines. This process results in the inhibition of T-lymphocyte activation and proliferation. Saclorin is primarily used to prevent organ rejection in patients receiving kidney, liver, or heart transplants and to treat various autoimmune conditions such as rheumatoid arthritis and psoriasis.
