Overview
Sacubitril + valsartan is an angiotensin receptor-neprilysin inhibitor (ARNI) developed by Kopran as a generic formulation for the treatment of heart failure. It consists of the neprilysin inhibitor sacubitril and the angiotensin II receptor blocker (ARB) valsartan. Sacubitril acts by inhibiting the enzyme neprilysin, which is responsible for the degradation of atrial and brain natriuretic peptides, thereby enhancing the beneficial effects of these peptides, such as vasodilation and natriuresis. Valsartan selectively blocks the AT1 receptor, inhibiting the vasoconstrictor and aldosterone-secreting effects of angiotensin II. This dual mechanism of action helps to reduce the strain on the heart and improve outcomes in patients with chronic heart failure with reduced ejection fraction (HFrEF).
