SAF-248 is a novel, potent, and highly selective small molecule inhibitor of Phosphoinositide 3-kinase delta (PI3Kδ), developed by Chongqing Fochon Pharmaceutical. It exhibits significant selectivity for the delta isoform over other PI3K isoforms (alpha, beta, and gamma), with an IC50 of 30.6 nM. In preclinical studies, SAF-248 demonstrated superior antiproliferative activity against a broad spectrum of B-cell lymphomas and leukemias, including diffuse large B-cell lymphoma (DLBCL), compared to the approved PI3Kδ inhibitor idelalisib. The drug targets the PI3K/AKT/mTOR signaling pathway, which is frequently dysregulated in hematologic malignancies. It is currently in the IND-enabling stage of development for the treatment of lymphoma.
