Overview
SAF312, also known by the brand name Libvatrep, is a potent, highly selective, noncompetitive antagonist of the human transient receptor potential vanilloid 1 (TRPV1) channel. It is being developed as a topical ocular analgesic in the form of eye drops for the treatment of ocular surface pain, including postoperative pain following procedures such as photorefractive keratectomy (PRK). SAF312 acts by directly blocking TRPV1 channels on sensory nerves in the cornea and conjunctiva, thereby inhibiting pain signaling triggered by various endogenous stimuli. Preclinical and early clinical studies have demonstrated that SAF312 has an acceptable safety profile with rapid absorption into ocular tissues and low systemic exposure. The drug does not exhibit anesthetic effects or delay wound healing after PRK.
